Capping Kit
This kit is designed for the in vitro enzymatic capping of mRNA. It allows the addition of a Cap0 or Cap1 structure on the 5’ terminus of mRNA, improving its stability and translation. The Vaccinia virus Capping Enzyme catalyzes the addition of a 7’methylguanylate cap to the 5’ end of mRNA (Cap0) while the 2’O-methyltransferase uses the Cap0-mRNA as substrate to produce a Cap1 mRNA by methylation of 2’OH of the first nucleotide using S-Adenosylmethionine (SAM) as methyl group donor.
Kit contents:
- Ribonuclease inhibitor (40u/µl), 50µl
- 2’O-Methyltransferase (50u/µl), 50µl
- Vaccinia Capping Enzyme (10u/µl), 50µl
- Capping Buffer 10X, 500µl
- GTP (10mM), 500µl
- SAM (32mM), 50µl
- Nuclease-free water, 2ml
Material needed not provided with the kit:
- mRNA to be capped
- 70% ethanol for RNA washing
- Nuclease-free buffers (1mM citrate buffer pH 6.4, TE buffer or RNAse-free water) for RNA resuspension.
- Materials or kits for purification of the RNA product
Storage: Upon receipt and for long-term use, store the kit at -20°C Kit’s components are stable for at least 1 year at the recommended storage temperature.
Shipping condition: The capping kit is shipped with dry ice.
