NanOZ LNP-siRNA

NanOZ LNP-siRNA are ready-to-use lipid nanoparticles (LNPs) loaded with small interfering RNA (siRNA) for gene silencing.

siRNAs induce RNA interference (RNAi), which leads to the sequence-specific degradation of the target mRNA. siRNA-mediated knockdown is transient, non-integrative and acts post-transcriptionally, which minimizes the risk of genomic alterations. This is relevant for stem cell research, functional genomics and therapeutic development. As a small, negatively charged molecule, siRNA requires a delivery vehicle for cellular uptake and protection from degradation.

NanOZ LNPs are lipidic spherical vesicles formed by four main components: ionizable cationic lipid, helper phospholipid, cholesterol and pegylated lipid, each having a distinct function. The saRNA self-assembles with the ionizable cationic lipid through electrostatic interactions. Formulation by microfluidic technology results in monodisperse LNPs with a narrow size distribution and an encapsulation efficiency >80%.

The range includes siRNA targeting GFP or GAPDH, designed to assess gene silencing efficiency, and a Cy5-labeled non-targeting siRNA (NT-Cy5), a control for siRNA delivery and biodistribution. The three siRNAs are also available in lipid nanoparticles formulated with the ionizable lipid D-Lin-MC3-DMA.
