NanOZ LNP-saRNA

NanOZ LNP-saRNA are ready-to-use lipid nanoparticles (LNPs) loaded with self-amplifying RNA (saRNA).

Self-amplifying RNAs encode a replicase that can copy the RNA after delivery into the cell, and are designed to produce a high level of protein expression over an extended period. Because of their size and negative charge, saRNAs need a delivery vehicle to enter cells and resist degradation.

NanOZ LNPs are lipidic spherical vesicles formed by four main components: an ionizable cationic lipid, a helper phospholipid, cholesterol and a pegylated lipid, each having a distinct function. The saRNA self-assembles with the ionizable cationic lipid through electrostatic interactions. Formulation by microfluidic technology results in monodisperse LNPs with a narrow size distribution and an encapsulation efficiency >80%.

The range currently includes three saRNA-LNPs: a GFP (eGFP) saRNA-LNP, to assess transfection efficiency and expression kinetics by fluorescence; a firefly luciferase (FLuc) saRNA-LNP, to assess expression kinetics by bioluminescence; and an ovalbumin (OVA) saRNA-LNP. Ovalbumin is a commonly used antigen for immunization and biochemical studies and an established model allergen for airway hyper-responsiveness.
